Streptavidin – R-Phycoerythrin
Streptavidin has a molecular weight of 53kDa and is purified from Streptomyces avidinii. It is a tetrameric protein composed of identical subunits. Each streptavidin monomer binds one biotin molecule with a very high affinity (dissociation constant (Kd) ≈10−14 mol/L) and specificity. The strength and specificity of the interaction has led it to be one of the most widely used affinity pairs in molecular, immunological, and cellular assays. This interaction can be detected by the fluorescent molecule.
R-PE is a 240kDa fluorescent protein from the phycobiliprotein family that is isolated from red algae. R-PE has a bright orange fluorescence with a high quantum yield, giving a brighter fluorescence than other conventional fluorophores and phycobiliproteins. The absorbance spectrum of R-PE covers a broad range of excitation wavelengths, and its emission maximum is at 575nm. The broad range of excitation wavelengths provides an advantage for multi-colour immunofluorescent staining or cell sorting.
